چکیده

I discovered the green fluorescent protein GFP from the jellyfish Aequorea aequorea in 1961 as a byproduct of the Ca-sensitive photoprotein aequorin (Shimomura et al., 1962; Johnson et al., 1962), and identified its chromophore in 1979 (Shimomura, 1979). GFP was a beautiful protein but it remained useless for the next 30 years after the discovery. My story begins in 1945, the year the city of Nagasaki was destroyed by an atomic bomb and World War II ended. At that time I was a 16-year old high school student, and I was working at a factory about 15 km northeast of Nagasaki. I watched the B-29 that carried the atomic bomb heading toward Nagasaki, then soon I was exposed to a blinding bright flash and a strong pressure wave that were caused by a gigantic explosion. I was lucky to survive the war. In the mess after the war, however, I could not find any school to attend. I idled for 2 years, and then I learned that the pharmacy school of Nagasaki Medical College, which had been completely destroyed by the atomic bomb, was going to open a temporary campus near my home. I applied to the pharmacy school and was accepted. Although I didn’t have any interest in pharmacy, it was the only way that I could have some education. After graduating from the pharmacy school, I worked as a teaching assistant at the same school, which was reorganized as a part of Nagasaki University. My boss Professor Shungo Yasunaga was a gentle and very kind person. In 1955, when I had worked for four years on the job, he arranged for me a paid leave of absence for one year, and he sent me to Nagoya University, to study at the laboratory of Professor Yoshimasa Hirata.

Transient expression of green fluorescent protein in radish (Raphanus sativus) using a turnip mosaic virus based vector

It is possible to use transgenic plants, as bioreactors, for the production of recombinant inexpensive chemicals and drug components. Transient gene expression is an appropriate alternative to stable transformation because it allows an inexpensive and rapid method for expression of recombinant proteins in plant tissues. In recent years, plant viral vectors have been increasingly developed as su...

A Model to Study the Phenotypic Changes of Insect Cell Transfection by Copepod Super Green Fluorescent Protein (cop-GFP) in Baculovirus Expression System

Background: Baculovirus expression system is one of the most attractive and powerful eukaryotic expression systems for the production of recombinant proteins. The presence of a biomarker is required to monitor transfection efficiency or protein expression levels in insect cells. Methods: The aim of this study was to construct a baculovirus expression vector encoding a copepod super green fluore...

Genetically Engineered Mesenchymal Stem Cells Stably Expressing Green Fluorescent Protein

Objective(s) Mesenchymal stem cells (MSCs) are nonhematopoietic stromal cells that are capable of differentiating into and contribute to the regeneration of mesenchymal tissues. Human mesenchymal stem cells (hMSCs) are ideal targets in cell transplantation and tissue engineering. Enhanced green fluorescent protein (EGFP) has been an important reporter gene for gene therapy. The aim of this stu...

A New Reporter Gene Technology: Opportunities and Perspectives

The paper summarizes the current status of the reporter gene technology and their basics. Reporter gene technology is widely used to monitor cellular events associated with gene expression and signal transduction. Based upon the splicing of transcriptional control elements to a variety of reporter genes, it “reports” the effects of a cascade of signaling events on gene expression inside cells. ...

Highly Efficient Transfection of Dendritic Cells Derived from Esophageal Squamous Cell Carcinoma Patient: Optimization with Green Fluorescent Protein and Validation with Tumor RNA as a Tool for Immuno-genetherapy

This study was conducted to optimize a highly efficient mRNA transfection into dendritic cells (DC) derived from esophageal squamous cell carcinoma (ESCC) patients. Applying an electroporation technique, in vitro synthesized Green Fluorescent Protein (GFP) mRNA was transfected as an indicator into the DCs derived from a healthy donor. Flow cytometry revealed 84.9% transfection efficiency for DC...

Cap analog and Potato virus A HC-Pro silencing suppressor improve GFP transient expression using an infectious virus vector in Nicotiana benthamiana

Transient expression of proteins in plants has become a choice to facilitate recombinant protein production with its fast and easy application. On the other hand, host defensive mechanisms have been reported to reduce the efficiency of transient expression in plants. Hence, this study was designed to evaluate the effect of cap analog and Potato virus A helper component proteinase (PVA HC-Pro) o...
